SUMMARY:

This research aimed to develop a mouthwash containing cetylpyridinium chloride (CCP) for dental caries prophylaxis, using direct compression technology. Therefore, the analysis of pharmaceutical excipients that could be used for this purpose and evaluation of the formulations characteristics was carried out. A pre-formulation study was carried out to assess the compatibility of the active pharmaceutical ingredient (API) with the excipients used, sorbitol, xylitol, menthol and magnesium stearate, through thermoanalytical techniques; differential scanning calorimetry (DSC), thermogravimetry (TG) and differential thermal analysis (DTA); and near-infrared (FT-NIR) and mid-infrared (FTIR) spectroscopy. The pharmacotechnical characteristics of the mixture of components in powder form and organoleptic and physicochemical characteristics of the lozenges obtained by direct compression were also evaluated. The DSC, TG and DTA results point to interactions between API and menthol, with changes in the API melting point; the data obtained by FT-NIR and FTIR demonstrated a physical and non-chemical interaction. A product with adequate pharmacotechnical characteristics regarding flow and compressibility was obtained, considering characteristics of Carr Index (CI), Hausner Ratio (RH), particle size, flow velocity, angle of repose, humidity and pH value analysis for evaluation. The lozenges obtained after compression met the desired characteristics, considering weight, hardness, thickness, friability, disintegration and organoleptic characteristics for evaluation. Thus, it can be considered that CCP lozenges with adequate pharmacotechnical characteristics were obtained, however with the use of ingredients that can interact with the API.
