Interleukin-17A (IL-17A) is the main cytokine synthesized by Th17 cells, its action is
mediated by interaction with the interleukin-17A receptor (IL-17RA). In the context of
renal transplantation, the presence of IL-17A in microenvironment renal and high levels
in the systemic circulation has been associated to the development of rejection. Given
this, genetic variations in the genes of IL-17A and IL-17RA may play a significant role
on the outcome of renal transplantation. Few studies have evaluated this role. Thus, the
aim of the present study was to investigate the association of single nucleotide
polymorphisms in the genes of IL-17A and IL-17RA with biochemical parameters of
renal function and with the allograft outcome in kidney transplant (KT) patients. Were
enrolled 148 KT patients and 175 healthy bone marrow donors (normal controls).
Among the 148 KT patients recruited, 111/148 (74.3%) had no rejection and the
remaining 37 (25.7%) exhibited rejection during transplantation. IL17A (rs2275913) and
IL17RA (rs4819554) gene polymorphisms were evaluated by PCR-RFLP method. Urea,
creatinine and glomerular filtration rate (GFR) were used as renal function parameters.
Women with rs2275913-AA genotype compared to women with rs2275913-GA or GG
genotype had 5.82 times more likely to be present in the transplanted group than in the
group of healthy controls (P = 0.018). There was decreased renal function (high values
of serum urea and serum creatinine and low GFR) of KT patients carrying the
rs2275913-A allele, as well as in carriers of the rs4819554-G allele in different periods
after transplantation. In summary, polymorphisms of the promoter region of IL17A and
IL17RA genes play an important role in decreasing renal function of renal transplant
individuals.
