SUMMARY:

Medulloblastoma (MB) is a pediatric type of brain cancer that affects the cerebellum. The current consensus is that MB can be divided into four separate subgroups: WNT, SHH, group 3 and group 4. Here, we inferred the transcriptional regulatory network and identified a set of master regulators (MRs) for each examined subgroup. WNT was not inspected, since it represents a minute portion of all MB cases and its patients show a 5-year survival rate close to 100%. A set of differentially methylated (DM) regulons was also determined for every subgroup. Afterwards, regulons simultaneously identified as MRs and DM regulons, for all investigated subgroups, were selected and named "regulons of interest". Hypergeometric testing was applied to validate the statistical significance of this group of regulons, obtained by intersecting all six sets generated by previous inspections. The test returned an adjusted p-value of 7.558e-24. The influence of the regulons of interest on other types of cancer was discussed, along with the future work required for a better understanding of their roles on the development of medulloblastoma.
