SUMMARY:

Alzheimer's disease is the most common cause of dementia in the elderly population. Described in the 20th century by the German doctor Alois Alzheimer, it has no cure and is a pathology of multifactorial etiology in sporadic cases, responsible for more than 90% of cases. Animal models with mutations in genes associated with familial cases of Alzheimer's have emerged intending to help to better understand which cellular and molecular mechanisms lead to the onset of this disease. However, it is not only Alzheimer's that presents clinical symptoms such as dementia, but other diseases such as Progressive Supranuclear Palsy can also lead patients affected to this clinical condition. Both Alzheimer's and Progressive Supranuclear Palsy are included in the tauopathies group; and Alzheimer's is still an amyloid disease, with an accumulation of β-amyloid protein in brain tissue. We seek in this work to investigate the possible differences between Alzheimer's and Progressive Supranuclear Palsy with integrative analysis at the genetic level and transcripts, to try to understand if there are advantages in this approach. The results were compared with animal models (5XFAD and TauD35). Thus, we find in the groups of patients with Alzheimer's a greater amount of differentially expressed genes concomitantly with genes having also altered isoforms, a more frequent process in Alzheimer's when compared to the group of patients with Progressive Supranuclear Palsy, which may show greater participation of events alternative splicing in Alzheimer's. Animal data have shown that even with the mutations, there is not much involvement of the splicing machinery in these models.
