A NOVEL THERMORESPONSIVE IBUPROFEN-LOADED NANOGEL-BASED APPROACH AS ANTIINFLAMATORY AGENT IN TOOTH BLEACHING: AN IN VIVO STUDY

SUMMARY:

The present study aimed to develop a nanogel containing ibuprofen, and evaluated the in vivo bioadhesive capacity, penetrability in the tooth surface submitted to bleaching and anti-inflammatory potential in the dental pulp of Wistar rats. Low energy technique and a copolymer with thermoresponsive properties, Poloxamer 407 (Pluronic F127®), were used to obtain nanogels from nanoemulsions. Average droplet size, polydispersity index (PdI) and zeta potential (ZP) were evaluated. The system maintained nanometric characteristics (< 150 nm), negative ZP and monodispersed PdI during the study, with 50% drug release within 6 hours. The animals were divided into the following groups: 1-control (CTRL); 2-bleached (BLE) (37.5% H2O2 concentration, Pola Office Plus®, SDI); 3-ibuprofen hydrogel (IBF-GEL) and; 4- ibuprofen nanogel (IBF-NEG). Scanning Electron Microscopy (SEM) and Confocal Laser Scanning Microscope (CLSM) analysis were performed after 24 hours. For CLSM analysis, 0.1% rhodamine B was added to the ibuprofen hydrogel and nanogel. The groups were analyzed in 24 hours and 14 days, and hemi-arches (N=10 hemi-arches/group) were processed for morphological and immunohistochemical analysis. SEM analysis showed a uniform residual layer formation, referring to the ibuprofen nanogel application on the tooth surface, and CLSM analysis revealed the ability to penetrate dental enamel and reach the pulp tissue. The IBF-NEG group showed the lowest inflammatory infiltrate scores and necrosis absence within 24 hours and 14 days. Inflammatory cytokines were more frequent in BLE group. Ibuprofen nanogel topical administration presents bioadhesion capacity, penetration into the dental structure and reduction of pulp inflammation generated by the bleaching treatment.
