RESUMO:

The sustainable use of natural resources has been a concern of the twenty-first century. The water is noteworthy since it guarantees the permanence of life on the planet. However the world faces the drinking water shortage crisis, which compromises access for that current and future generations. Arid and semi-arid regions are those most in need with a low water availability due to unfavorable environmental conditions. The reservoirs for human supply built in those regions are centres for the subsistence of the population, however an anthropic action is one of the main factors that promote the contamination of these water bodies. The analysis on the microbiological water quality has been intensified to the data survey on the distribution and ecology of organisms. Then comes the need for the microbiota identification studies that determines the quality of water. Improved techniques on identifying and sequence comparison appeared in the 70’s, among them the metagenomics, characterized as the genetic study of a given ecosystem which allows easy viewing on the microorganisms distribution and classification. In this context, this study aimed to identify and compare the microorganisms diversity present at Piancó-Piranhas-Assu River Basin, in the state of Rio Grande do Norte, using molecular techniques / metagenomics. On the sample analysis of Armando Ribeiro Gonçalves Dam, it was found the prevalence of bacteria dominium, followed of archaea, viruses and eukaryotes. Among the bacteria, for example, the highlighted filo was the cyanobacteria, which is related to the eutrophication phenomenon in dams. The study also aimed to identify in high school biology textbooks referenced in 2015 PNLD Biology Guide, the presence and type of didactic approach of DNA and biotechnology issues. It was observed that despite the need and importance of the CTS approach to biology teaching, most of the textbooks do not respect the scientific and technological knowledge with a direct implication on society and the environment. This work also aimed to characterize the use of teaching strategies in DNA matter carried by the local teachers. It was found that 73% of teachers have difficulty teaching topics related to DNA and 100% of them said that students also have difficulty in understanding. The analysis of the questionnaires leads to the conclusion that the use of some teaching resources does not exclude the possibility that the dominant model of education remains still the traditional, highlighting the importance, for example, of the continuing education courses. In addition, the preliminary results of this study and reflections from it, will lead to realization of continuing education courses through lectures and interventions with regard to the socialization of scientific knowledge on the dams’ microbiota and especially with molecular knowledge, the study of DNA and molecular techniques, which allowed access to such knowledge.
