RESUMO:

Reserpine induces failure in the storage capacity of monoamines in the synaptic vesicles, generating a depletion of these substances in the terminal nerve and increased oxidative stress (OE). Several evidences shows that this drug causes the appearance of motor and non-motor symptoms similar to those seen in Parkinson's disease, with its application protocol commonly performed during the light phase. Some studies, however, suggest that in the light phase there is a lower concentration of antioxidants in the body due to daily oscillations in the production of enzymes such as catalase and superoxide dismutase, in addition to the neurohormone melatonin, which present peaks during the dark phase. These oscillations, therefore, may represent a phase-dependent vulnerability to disorders resulting from damage to DNA structure, lipid peroxidation and protein oxidation. Many of these parameters of biological rhythmicity are not taken into account in the standardization of experimental models, which may distract the results obtained in these animals from the pathophysiology of some disorders. The aim of this work is to observe if the pharmacological model of reserpine in rats (Santos et al., 2013), when applied during the dark phase, has the same effect of induction of symptoms of PD compared with its standard protocol of application in the light phase. It will be used 160 animals with an approximate age of 3 months. The animals will be submitted to the administration of reserpine in two distinct phases, activity and rest; the effect of the drug will be assessed through the catalepsy test, in groups of addition or not of antioxidants and melatonin. Subsequently, neurochemical parameters related to the death of dopaminergic neurons, reactive astrogliosis and oxidative stress in the nigrostriatal pathway will be evaluated.
