RESUMO:

Parkinson’s disease (PD) is a chronic and progressive neurodegenerative disorder that mainly affects central dopaminergic cells and causes motor function deficits. Nowadays we know that PD includes some sort of non-motors symptoms like as sexual dysfunction, cognitive and olfactive deficits, depression, anxiety disorders, sleep disruption. These group of symptoms maybe be a consequence of dysfunctions in others neurotransmission systems like the cholinergic, serotoninergic and histaminergic systems. Animal models and human post-mortem studies reveal that PD background involves an increased histamine release,  number of histaminergic projections and receptors. This alterations has been correlated with the worsening of the motor and cognitive symptoms. Although, these studies do not extend the discussion about how that altered innervation can affect other symptoms. In this manner, here we propose to evaluate how a dopamine depletion changes histamine production and how this event can affect the circadian rest-activity rhythm of rats under 6-OHDA administration. Apart from this, we propose to observe how the rhythm alterations correlate with the expression of histidine decarboxylase, BMAL1 protein and tyrosine hydroxylase by immunocytochemistry and fluorescence labeling. Preliminary results show that 6-OHDA infusions (chronic or acute protocols) in the lateral ventricles are not able to induce alterations in many chronobiological parameters like %variance, amplitude, mesor, alfa (%Tau), intraday variability and acrophase. We cannot observe any motors dysfunctions in catalepsy test. In addition, we did not observe changes in tyrosine hydroxylase expression in the substantia nigra and dorsal striatum. Our perspective is that a new infusion protocol in the third ventricle can induce the behavioral outcome expected in PD.
