RESUMO:

Glutamate (Glu) is a fundamental neurotransmitter in the processing of sensory functions and in the development and maintenance of complex cognitive functions such as learning and memory. Among the encephalic regions to which such neurotransmitter can be found, stands out the thalamus, a structure directly involved with integrative and modulatory functions of the nervous system. Considering that the thalamocortical projection systems are fundamental for regulating sensory information and for integrating this information with the associative cortex, the present study aims to characterize the morphology of the thalamus nuclei of the rock cavy (Kerodon rupestris), a typical rodent of the Northeast of Brazil, as well as the glutamatergic neuronal population in these nuclei. For this, 5 young adult rock cavy will be used. After perfusion and microtomy stages (for 2 animals, so far), immunohistochemistry for Glu, Nissl technique and collection and analysis of images were performed in order to characterize the cytoarquiteture of thalamic nuclei, as well as morphometrically qualify the glutamatergic neuronal populations. Data were processed from one-way ANOVA, with Tukey post-hoc test and Kruskal-Wallis non-parametric test. The nuclei analyzed so far are of the midline and intralaminar of the thalamus. Immunohistochemistry for Glu demonstrated efficient labeling of glutamatergic cells reflecting morphological characteristics with variations between nuclei. Greater cell area trend was observed in the intralaminar nuclei, but without statistical inference as a consequence of reduced n. Regarding the distribution of the rostrocaudal cell area to nucleus, we found that among the nuclei of the midline, the paraventricular (PV) and intermediodorsal nuclei (IMD) present a predominance of greater area at caudal levels, rhomboid (Rh) and reuniens (Re) at the rostrais levels and the paratenial (PT) at medium levels. Among the intralaminars nuclei, the medial central (CM) and paracentral nuclei (PC) at caudal levels, central lateral (CL) and parafascicular nuclei (PF) at medium levels. Confirmation of the results will occur with the processing of other animals of study and subsequent data analysis. The morphological characterization of the glutamatergic fibers founded and the differences between the nuclei may allow the suggestion of different patterns of synaptic organization of this neurotransmitter besides confirming its great functional repertoire.
