First evidence of the presence of anti-Leishmania antibodies in domestic cats (Felis catus) in a city in northeastern Brazil endemic for human and canine leishmaniasis - an exploratory study

In domestic felines, the number of diseases caused by parasites of the genus Leishmania is
becoming increasingly significant, although the role of cats in this cycle is still unclear.
Leishmaniasis is an illness caused by the protozoan Leishmania spp., which affects humans,
wild and domestic animals, and is considered a zoonosis. Its vertebrate hosts and reservoirs are
domestic and wild dogs, although cases have been reported in other species, including domestic
cats (Felis catus). The parasite is vector-borne and transmitted through the blood meal of female
sandflies, with Lutzomya longipalpis being prevalent in Brazil. Previous studies in cats have
reported that these animals are affected by visceral leishmaniasis (VL) and can become infected
and develop the disease, exhibiting variable clinical signs. Consequently, most studies are
limited to dogs, and few focus on investigating Leishmania sp. in other mammals.The objective
of this research was to identify the seroprevalence of antibodies against Leishmania spp.
through the indirect ELISA test and establish a standardized in-house ELISA protocol using
antigens from Leishmania infantum, L. amazonensis, and T. cruzi in serological samples from
domestic felines from the sterilization outreach program conducted by the Vigilance and
Zoonosis Unit (UVZ) in Natal, located in the state of Rio Grande do Norte. A total of 130
domestic felines underwent clinical examination, and blood samples were collected from the
jugular or cephalic vein. The results of the in-house ELISA test indicated that 28 domestic
felines (21.54%) showed potential seroreactivity, 9 domestic felines (6.92%) demonstrated
non-seroreactivity, and 93 domestic felines (71.53%) were serologically inconclusive for anti
Leishmania infantum IgG. Due to the increasing number of reported cases of leishmaniasis in
animals in Brazil, particularly considering the endemicity of the Northeast region, further
studies are necessary to improve and standardize ELISA tests for the diagnosis of feline
leishmaniasis. Thus, this study was able to demonstrate the involvement and exposure of felines
in Natal, Rio Grande do Norte, to this zoonosis and their potential participation in the
epidemiological chain of visceral leishmaniasis transmission.
