SUMMARY:
Species of the Ouratea genus belong to the Ochnaceae family and are found mainly in
Northeast Brazil. From the seed of its fruits, a sweet and aromatic fat can be extracted,
the bati butter, rich in unsaturated fatty acids, used in the frying of foods and popularly
as a healing and anti-inflammatory. However, its physicochemical characteristics and
bioactive properties are poorly studied. But the ucuuba butter, extracted from the
ucuubeira (Virola Surinamensis), found in the river zone of the Amazon, is fat rich in
saturated fatty acids. Its characterization and bioactive properties are not widely described
in the literature. Thus, the objective of this study was to evaluate the physicochemical and
bioactive properties and the cytotoxicity of bati and ucuuba butters. Initially, the
determination of the fatty acid profile and the physicochemical characterization of the
samples were carried out. Furthermore, the samples were analyzed by FTIR (Fourrier
transform infrared) and 1H NMR (hydrogen nuclear magnetic resonance). The
antioxidant activity was evaluated by capturing the DPPH (2,2-diphenyl-1-picryl
hydrazyl) and ABTS (2,2'-azinobis 3-ethylbenzothiazoline-6-sulfonic acid) radicals, the
profile of the total phenolic components by the method Folin-Ciocalteau and cytotoxic
activity through the colorimetric assay of MTT ([3-(4,5-dimethylthiazol-2yl)-2,5-
diphenyl tetrazoline bromide]). As for the lipid profile and the physicochemical
characteristics, the bati butter presented 66.06% of unsaturated fatty acids, with linoleic
acid in greater proportion (45.36%). The bati butter, therefore, had a higher iodine content
(23.72 cg I2.g -1) than ucuuba butter (5.33 cg I2.g -1). In ucuuba butter, there was a higher
percentage of saturated fatty acids (85.75%), with emphasis on lauric acid (37.80%). The
hydrophilic fraction of butters was used for quantification of total phenolic compounds
(CFT) and evaluation of antioxidant activity in vitro. The bati butter presented a CFT of
76.57 mg EAG.kg -1 and the ucuuba butter of 76.25 mg EAG.kg -1 . The hydrophilic
fraction of ucuuba butter was highlighted in the in vitro antioxidant capacity in the ABTS
radical capture assay, presenting an IC50 (concentration in which 50% of the radicals
were inhibited) of 2.58 mg/mL. In addition, the two butters and their respective
hydrophilic and lipophilic fractions did not show cytotoxic activity for cell lines CHO
K1 (Chinese hamster ovary cells) and HepG2 (human hepatocarcinoma cell line), with
cell viability greater than 70%. in all samples. As it has a rich composition of unsaturated
fatty acids and does not present cytotoxicity, it is possible to suggest the exploitation of
bati butter by the food industry. On the other hand, ucuuba butter with important
antioxidant activity, with a predominance of lauric acid, a fatty acid with surfactant and
emollient properties, reveals itself as a raw material of potential interest for the cosmetic
and pharmaceutical industry.
