SUMMARY:
The plant species Commiphora leptophloeos (Mart.) J.B. Gillett, stands out for being a
medicinal plant used in the treatment of inflammation in general. Therefore, this work aims to
evaluate the antinociceptive activity and subchronic toxicity of the hydroethnolic extract of the
stem bark of C. leptophloeos in mice. Initially, subchronic toxicity was evaluated for 28 days at three
different doses (200, 400 and 800 mg/kg) orally and behavioral analyzes were performed (Hippocratic
screening, Campo Aberto test and Rotarod), mortality, alteration of hematological parameters,
alterations macroscopic and histopathological analysis of organs (heart, liver, spleen and kidneys), and
evaluation of markers of inflammation and oxidative stress (MPO and MDA).The potential
antinociceptive effect of the EHCL (100, 200 and 400 mg/kg) was evaluated in vivo through
models of pain induced by chemical and thermal agents, such as the abdominal writhing tests
induced by acetic acid, formalin and hot plate, respectively. In order to evaluate the participation
of opioid receptors, ATP-sensitive K+ channels and the L-arginine-NO pathway in the
antinociceptive action, pre-treatment with naloxone, glibenclamide or methylene blue was
performed, respectively, before the administration of the dose of 400 mg/kg. The subchronic
toxicity study showed that the oral extract at doses of 200, 400 and 800 mg/kg did not present
significant changes in behavioral, locomotor and hematological parameters. However, on
histological analysis, the dose of 800 mg/kg showed signs that indicate nephrotoxicity. The
EHCL at doses of 100 and 200 mg/kg showed an antinociceptive effect against chemical
stimuli, while the dose of 400 mg/kg showed an antinociceptive effect against chemical and
thermal stimuli. Glibenclamide associated with EHCL partially reduced the antinociceptive
effect of the extract, demonstrating possible involvement with ATP-sensitive K+ channels. The
administration of methylene blue with EHCL caused a potentiation of the antinociceptive effect
of the extract, indicating involvement in the cGMP pathway. The use of naloxone together with
the extract resulted in the inhibition of the analgesic effect of EHCL, demonstrating that the
extract is involved in the opioid system. Finally, the results obtained in this work justify the use
of C. leptophloeos stem bark in folk medicine to treat pain and contribute to the development
of a possible safe and effective herbal medicine with analgesic properties.
