SUMMARY:

Alzheimer's disease (AD) is characterized by a neurodegenerative disorder that causes irreversible memory loss, being the main cause of dementia in the elderly population. The pathology is not fully understood, but it has as main causes some metabolic alterations that result in the characteristic signs of the disease. Including deposition of beta-amyloid protein plaques and formation of neurofibrillary tangles in the hippocampus region and in the cerebral cortex, in addition to sympathetic dysfunction with reduced cholinergic activity. Currently, the therapeutic arsenal of this disease is limited to some substances that act in the inhibition of the cholinesterase enzyme that blocks the effects of acetylcholine reduction, resulting in a neuroprotective effect. This therapy only has the ability to improve the quality of life, but it does not prevent the progression of the disease. Therefore, new options for natural products become interesting due to the great biodiversity of medicinal plants found in the country, mainly in the Caatinga biome exclusive only in the Brazilian territory. Erythrina velutina has established popular therapeutic use and has isoquinoline alkaloids in its composition that have demonstrated psychoactive activities in pilot experiments. Thus, the objective of the work is to evaluate the biological potential in vivo of fractions enriched in E. velutina seed alkaloids using zebrafish (zebrafish, Danio rerio) as an experimental model. The seeds were crushed and submitted to hydroethanolic extraction by static maceration, after the filtration and drying of the extract, an acid-base extraction was performed to obtain the fraction enriched in alkaloids. Chromatographic analyzes in thin layer using mobile phase chloroform / methanol with concentration gradient (100 of chloroform; 7: 3, 5: 5; 3: 7 and 100% methanol) and in gas chromatograph coupled to the mass spectrometer for purity analysis and structural characterization with Helium as carrier gas and The temperature program was: 100 ° C for 0 min, 220 ° C for 10 min and 290 ° C for 11 min. The alkaloids erysodine and erisovine were tentatively noted. This sample will be used in biological tests in zebrafish to assess the bioactive potential in the face of aspects related to Alzheimer's disease, such as cognition and memory. At the end of this work, we seek to contribute to the understanding of the biological effects in vivo of benzylisoquinoline alkaloids present almost exclusively in species of the genus.
