Ferulic acid (FA) is a phytochemical constituent belonging to the polyphenois group found in vegetables, mainly in cereals like rice bran and maize, with emphasis on its photoprotective and antioxidant activity. Thus, the FA and its structurally related compounds have been widely studied for application in photoprotective and anti-aging cutaneous formulation. In the development of topical formulations, the in vitro cutaneous release assays are a fast, low-cost test that enables preliminary results. However, the use of FA is limited by the instability of the molecule against light and/or oxidation. As a result, its stability can be improved by the FA complexation with cyclodextrins, dispersion in polymers or inclusion in cyclodextrins followed by the association of a polymer forming a multicomponent system. In the light of this context, the objective was to evaluate the interactions in binary mixtures (1:1) between FA, cyclodextrins and polymers. Furthermore, it was intended to obtain and characterize a biomimetic membrane model that is similar to the lipid composition of the skin, as well as to apply this membrane in the evaluation of the FA release from cosmetic formulations. The binary mixtures were submitted to analysis by differential scanning calorimetry (DSC), Thermogravimetry (TG), X-ray diffraction (XRD), Fourier transform infrared spectrophotometry (FTIR) and Scan electron microscopy (SEM). The biomimetic membranes were obtained by impregnation of a lipidic solution in synthetic membrane and characterized by DSC, atomic force microscopy (AFM) and SEM. gel and gel-cream formulations containing FA 0.5% (w/w) were evaluated using the biomimetic membrane through Franz diffusion cells. Thermoanalytical analysis showed that polymers and cyclodextrins provided thermal protection for FA. Through the diffractograms it was possible to observe that cyclodextrins have granted amorphous behavior to the molecule and the polymers reduced the intensity of crystalline reflections. The spectra of the binary mixtures showed predominance of physical interactions. In relation to the biomimetic membrane, it was possible to verify the impregnation of the lipidic solution in the polymer mesh of the synthetic membrane. In the experimental conditions evaluated, it was found that the gel formulation released a greater amount of PA in a shorter time interval, while the gel-cream formulation presented a lower and slower release of the active. At the end of the study, it was possible to observe that cyclodextrins and polymers increased the PA stability and reduced its crystallinity, with potential use in methods aimed at molecule’s solubility increase and stability . In relation to the biomimetic membrane, greater resistance was observed for PA diffusion, thus provoking distinct release behaviors among the formulations. The same result was not observed in the release using the synthetic membrane, where the formulations presented the same release profile.
