RESUMO:

Accidents with venomous animals, mainly snakes and scorpions, are considered a serious public health problem. In Brazil, ophidian accidents are mainly provoked by snakes of the genus Bothrops, and those with scorpions, by the species Tityus serrulatus. The conventional treatment is the use of antivenom serotherapy, which in addition to presenting some limitations, such as difficult access in some regions and high cost, is only able to neutralize the poison toxins, being inefficient in inhibiting the endogenous reactions triggered by their presence. These reactions are related to the major poison damage in the organism, such as the pulmonary inflammatory response to the scorpion venom and the tissue necrosis to the bothropic venom. Due to these limitations, several research groups have been studying and demonstrating the benefits of alternative therapies with plant species. For this purpose, they are based mainly on folk medicine and on the fact that these plants are a rich source of pharmacologically active compounds and inhibitors; its use has as main advantages the easy accessibility, the low cost and, mainly, the potential to diminish the endogenous inflammatory response as well as some local effects, such as formation of oedemas, hemorrhage and tissue necrosis, that are weakly neutralized by the serotherapy. In this context, the aim of this paper was to perform a phytochemical analysis of the fruit juice of Hancornia speciosa and its ability to inhibit the effects of the venom of the scorpion Tityus serrulatus and the snake Bothrops jararaca. The phytochemical analysis was performed using high performance liquid chromatography MS / MS, and the anti-corticosteroid activity was evaluated following a treatment protocol: administration of the venom via subcutaneous injection in Swiss mice, euthanasia of the animals two hours later with excess of xylazine/ketamine; afterwards, collection of lungs, kidneys and blood. The following analyzes were performed with the lungs: quantification of IL-1β cytokines, IL-6 and TNF-α, concentration of Evans blue, myeloperoxidase enzyme (MPO) and relative lung weight percentages; with the kidneys, nitrite dosage and lipid peroxidation; and hematological and biochemical parameters were evaluated with the blood. In relation to the bothropic venom, it was evaluated the capacity of the juice to inhibit its edematogenic activity - using the mouse paw edema model - and its myotoxic activity - through the dosage of the enzymes creatine kinase and lactate dehydrogenase and through the indirect method of quantification of hemoglobin in the muscle. Finally, the inhibition of the dermonecrotic lesion was evaluated. The results obtained in relation to phytochemistry showed the presence of thirteen compounds: chlorogenic acid, isochlorogenic acid, 3-feruloylquinic acid, ramnetin, rutin, 5-feruloylquinic acid, quercetin-3-O-hexoside, kaempferol rutinoside, kaempferol-hexoside and quercetin. Observations regarding the inhibition of antiscorpionic effects demonstrated that the Hancornia speciosa juice was able to decrease some important parameters such as the cytokine production, the myeloperoxidase activity, the concentration of Evans blue on the lung tissue, the lipid peroxidation and the nitrite dosage. In relation to the bothropic effects, the juice was able to significantly inhibit the dermonecrotic lesion, the edematogenic activity and the concentrations of the enzymes creatina kinase and lactate dehydrogenase. Through the results, it can be concluded that the juice of Hancornia speciosa Gomes has great potential to be used as an adjuvant in the treatment of anti-corticosteroid and antibothropic poisonings.
