RESUMO:

Prosopis juliflora (PJ) is an invasive plant of the Brazilian Caatinga and presents characteristics in its metabolic composition of interest in cosmetic products as moisturizing and anti-wrinkle actives. This work aims the development of cosmetic formulations in the form of a solid core containing PJ extract that upon contact with water will form instantly a gel for single dose use. It also aims the in vitro and in vivo safety study of the extracts obtained and the clinical efficacy of the moisturizing and anti-wrinkle activities. The fruit (also known as pods) development and optimization of the extraction process PJ was conducted from statistical experimental design changing the temperature and extraction time. To select the best conditions the variables as yield, total sugar content and total antioxidant activity were considered. In addition, the presence of phenolic compounds and proteins were also evaluated. The in vitro safety assessment through the cytotoxicity test using the neutral red colorimetric method was performed. The samples were purified by centrifugal ultrafiltration and gel filtration chromatography, followed by acid hydrolysis and analysis of the monosaccharide composition by paper chromatography and nuclear magnetic resonance. Finally, the preformulation study, flow and compressibility analysis of the formulations was carried out. The statistical experimental design showed that the best extractive conditions are those with the lowest time and temperature named PJ1 (40ºC / 2h), PJ2 (34ºC / 3,5h) and PJ3 (25ºC / 3,5h). However, the PJ2 sample was chosen for the continuity of the study and the samples were considered non-cytotoxic. The fraction containing the metabolites with molecular weight greater than 3KDa of PJ2 presented a single peak, probably composed mostly by glucose and galactose, mannose and arabinose traces, later characterized as an α-glucan by nuclear magnetic resonance. In the pre formulation study, the excipients were evaluated for solubility and functionality. Eight formulations were delineated using mannitol as diluent, magnesium stearate and talc as lubricants and glidants, different disintegrating agents (croscarmellose sodium and sodium glycolate) and viscosity donors (carboxymethyl cellulose and sodium polyacrylate). The formulations showed excellent flow and compressibility suitable parameters, required for the compression process of the solid cores.
