RESUMO:

Cutaneous leishmaniasis affects millions of people every year, especially in poor and tropical countries, where there are not investment and development of new drugs. In addition, the drugs currently used generate high toxicity and numerous adverse effects, making it necessary to develop new alternatives drug. Nanotechnology has been outstanding for presenting efficient solutions in the development of new drugs. For this reason, the objective of this work was to incorporate the oil fraction of annatto (Bixa orellana L.), a bioactive compound with proven anti-leishmania activity, into nanostructured lipid carriers. The nanoparticles were produced by the fusion-emulsification method and were characterized in terms of particle size, polydispersity index, morphology, zeta potential, 90-day storage stability, encapsulation efficiency, thermal stability, crystalline profile, lipid peroxidation, anti-leishmania activity and the evaluation of influence of 3% (w/w) of synthetic Laponite® clay on the system. The assays showed the effectiveness of the method with nanoparticles ranging from 173.73 nm ± 1.15 to 185.23 nm ± 1.80, polydispersity less than 0.3 and zeta potential between -23.07 mV ± 0.15 and -39.13 mV ± 0.50 and stability maintained for 90 days, with no variation due to the presence of clay. Carriers remained stable when submitted to body temperature (32ºC - 37ºC), with mass loss at approximately 100ºC. The crystalline profile of nanostructured lipid carriers favored the encapsulation of the oil, with encapsulation efficiency of 99%. The presence of Laponite® reduced the crystallinity of the nanostructures, favoring the packaging of the oil. The already known anti-oxidant activity of annatto was demonstrated by reducing the formation of malondialdehydes, from 10.20 μM ± 2,64 in the formulation that did not contain oil to 2.64 μM ± 0.91 in which the oil was present. The carriers showed anti-leishmania activity similar to amphotericin B and greater than Glucantime® and unencapsulated annatto oil. These results demonstrate a stable system with proven biological activity that should be seen as a potential therapeutic alternative for cutaneous leishmaniasis and, therefore, requiring further studies and investments.
