RESUMO:

Kalanchoe brasiliensis Cambess and Kalanchoe pinnata (Lamarck) Persoon (Crassulaceae),
popularly known as saião and coirama, are widely used in traditional medicine in the
treatment of peptic ulcers and inflammation. It is worth mentioning that K. pinnata is present
in RENISUS (2009). In this context, the objective of the present work was to characterize the
chemical markers present in the leaf juices of both species and to evaluate the antiulcer and
topical anti-inflammatory activity. Phytochemical characterization was performed by Thin
Layer Chromatography (TLC) and Ultra Low Efficiency Liquid Chromatography Coupled to
Mass Spectrometer (HUPLC-MS). The antiulcer activity was evaluated in the ethanol and
indomethacin induced ulcer models, and the total acidity, pH and volume were evaluated in
the pylorus ligature model, both in Wistar rats. Juice at doses of 125, 250 and 500 mg/kg and
ranitidine (50 mg/kg, orally) were given orally 24 and 1 hour prior to induction. The topical
anti-inflammatory activity was evaluated in the carrageenan-induced paw edema model and
croton oil-induced ear edema in Swiss mice, using dexamethasone (1mg/g, topical) as the
standard drug. The TLC analysis revealed the presence of flavonoid stains in the juices of the
two species, after revealing with the Natural Reagent A and allowed to observe that the
species in question have a different flavonoid profile. In the analysis by HUPLC-MS the juice
of the leaves of K. brasiliensis presented glycosylated flavonoids derived mainly from
patulelet, while that of K. pinnata showed mostly glycosylated flavonoids derived from
quercetin. The pre-treatment with the juice of the leaves of K. brasiliensis at doses of 250
mg/kg (P <0.01) and 500 mg/kg (P <0.001) and juice of K. pinnata leaves at doses of 250 mg
/ kg (P <0.001) and 500 mg/kg (P <0.001) significantly reduced the lesions compared to the
positive control in the ethanol induction model. In the indomethacin induction model, the
juice of the leaves of K. brasiliensis also presented significant results at doses of 250 (P
<0.05) and 500 mg/kg (P <0.01) and K. pinnata at doses of 250 P <0.01) and 500 mg/kg (P
<0.001). The reduction of the lesions was accompanied by reduction of oxidative stress,
observed by the increase of the total content of glutathione and the reduction of
malondialdehyde levels. In addition, inflammatory markers were reduced: myeloperoxidase,
IL-1β and TNF-α. It was also possible to observe a cytoprotective effect from histological
evaluation and reduction of iNOS expression by immunohistochemistry. In the pylorus
ligation model, leaf juices from both species did not change the acidity, pH and volume of the
gastric juice. In the ear edema model, the formulations containing the three juice
concentrations of the leaves of K. brasiliensis significantly reduced the edema when
compared to the placebo group (1.25% P <0.05, 2.5% P <0.01 And 5% P <0.01). Only the
formulation containing the juice of the K. pinnata leaves at 5% concentration presented a
significant result (P <0.01). In the paw edema model, the formulations containing the juice of
the leaves of K. brasiliensis at concentrations of 1.25 and 2.5% significantly reduced (P
<0.05) the edema in the time 4 h. The formulation at 5% concentration significantly reduced
edema at 1 h (P <0.001), 2 h (P <0.01), 3 h and 4 h (P <0.01). Regarding the formulations
containing the juice of the K. pinnata leaves, the concentration of 1.25% significantly reduced
edema in time 1h (P <0.01) and in time 2h (P <0.05), in the concentration of 5% significantly
reduced in time 1h (P <0.05). The decrease in edema was followed by reduction of the
activity of the enzyme miloperoxidase. Thus, it can be concluded that leaf juice of both
species and study showed antiulcer and topical anti-inflammatory activity in in vivo models,
results that justify the popular use of the species.
