RESUMO:

The development of new drugs is an activity of great importance for the
pharmaceutical industry and involves several steps, among them the quality
and safety monitoring of the substances of interest. P-coumaric acid, via an
esterification process, gives methyl p-coumarate, ethyl p-coumarate and
isopropyl p-coumarate, which are targets of several biological studies, but
information about its physicochemical characteristics, stability and analytical
methodology for its identification and quantification are scarce in the literature.
In this context, this work aims at the thermal and chromatographic evaluation of
these esters, aiming to evaluate their stability and physicochemical
characteristics contributing in the new drugs research and quality control of
drugs that will present these molecules as active principle. The analysis by
DSC, DTA and TG showed that the p-coumaric acid derivatives, despite their
structural similarity, have different thermal behavior, so that it is possible to use
these techniques to identify them. Methyl p-coumarate and ethyl p-coumarate
showed melting point at 131 ° C and 65 ° C, respectively, since the p-coumarate
isopropyl did not show a melting process. The three samples have good thermal
stability, with methyl p-coumarate being the most stable. A method for
simultaneous identification of these compounds by UHPLC was developed, the
method was also efficient in the identification of its degradation products when
applied in the study of forced degradation.
