RESUMO:

Genipa americana Linneus is a fruit tree popularly known as “Jenipapo”, from the Rubiaceae Family, native from Amazonia, distributed throughout both Central and South Americas. The species has a recognized use in folk medicine, in which it is used to treat various conditions (e.g. diarrhea, anemia and liver disease). However, there are still few phytochemical and pharmacological studies that support its use by the population. Provided that, the present paper focuses on the characterization of the chemical markers present in the leaves of G. americana, using chromatographic techniques and thermal analysis, as well as evaluating its anthelmintic and antifungal activity. The hydroethanolic extract of the leaves (HEL) was prepared by maceration in hydroethanolic environment at 70%, this was partitioned with organic solvents in order of increasing polarity (Petroleum ether, chloroform, ethyl acetate, and n-butanol). The fractions that have higher concentrations of the compounds of interest were selected to follow up the isolation steps. The evaluation of the phytochemical profile of the leaves was performed preliminarily by TLC (Thin-layer Chromatography) and then by UHPLC and LC-MS. A UHPLC characterization method is being developed and optimized, something innovative and not yet described in the researched literature for this specie. Through TG (Thermogravimetry) and DSC (differential scanning calorimetry) it was possible to identify and select the thermal events applicable to characterisation of the stratum of the leaves (EHF), the '' TF '' (residue of maceration), of remacerado and of the fractions of the partition (Fr.ETER, Fr.CHCl3, Fr.AcOEt, Fr.BuOH e Fr.Residual). Through the process of isolation by chromatographic techniques it was possible to obtain flavonoids and iridoides, which are in a phase of structural elucidation. By means of HPLC in, it was possible to identify two iridoides: 1-hidroxi-7-(hidroximetil)-1H,4aH,5H,7aH-ciclopenta[c]piran-4 carbaldeído, present in EHF and in all the fractions analyzed; and 7-(hidroximetil)-1-metoxi-1H,4aH,5H,7aH-ciclopenta[c]piran-4-carbaldeído, present only in the fraction Fr.CHCl3. After identification of the compounds of interest, the method will be validated according to the rules of the RDC No. 899/2003 of ANVISA and international standards.
