RESUMO:

The species Kalanchoe brasiliensis Cambess and Kalanchoe pinnata (Lamarck) Persoon
(Crassulaceae), popularly known as "Saião" and "Coirama", are used in traditional medicine
in the treatment of peptic ulcers and skin inflammations. Despite the wide popular use there is
no monograph that specifies the quality parameters of these plant species. It is worth
mentioning that K. pinnata is present in the National List of Medicinal Plants of Interest of
the Unified Health System - RENISUS (2009). In addition, there are no studies in the
researched literature that compare the local anti-ulcer and anti-inflammatory activity of leaf
juice of the two species. In this context, the present study aims to evaluate the anti-ulcer and
topical anti-inflammatory activity, as well as to characterize the biological markers present in
the juice of the leaves of the two species. For this, phytochemical characterization was
performed by Thin layer chromatography (TLC), which revealed the presence of flavonoid
bands in the leaf juice of the two species. By means of the preliminary analysis by TLC it was
possible to visualize that the juices of the leaves of the species K. brasiliensis and K. pinnata
present different chemical profile when comparing the coloration and Rf of the revealed
bands. The anti-ulcer activity of leaf juice of K. brasiliensis and K. pinnata was evaluated
using ethanol and indomethacin ulcer induction models, using ranitidine (50mg/Kg) as the
standard drug. The topical anti-inflammatory activity was evaluated in the carrageenan
induced paw edema model and croton oil-induced ear edema in Swiss mice, using
dexamethasone as the standard. In the evaluation of the anti-ulcer activity the juice of the
leaves of K. brasiliensis and K. pinnata and ranitidine were administered orally (125, 250 and
500mg/kg) 24 and 1 hour before the induction of the lesions. The pre-treatment with the juice
of the leaves of K. brasiliensis and K. pinnata revealed a significant improvement of the
gastric lesions, characterized by the reduction of the ulceration index in the two models of
ulcer evaluated. The reduction of the lesions was accompanied by reduction of oxidative
stress, observed by the increase of the content of total glutathione, an important antioxidant.
The juice of the leaves of K. brasiliensis and K. pinnata also significantly improved the
topical inflammatory process, reducing edema in the model of paw and ear edema, both of
which could have an effect on the reduction of the local inflammatory process. We emphasize
that preliminary data are necessary, and it is necessary to continue the phytochemical studies,
analysis by High performance liquid chromatography coupled to mass spectrometer (HPLCMS)
and pharmacological to deepen the mechanisms involved in the beneficial effects of
these compounds on gastric secretory function by means pylorus ligature, besides the
evaluation of prostaglandin E2 (PG2) levels, cytokines (TNF-α, IL-6 and IL-8),
myeloperoxidase (MPO), malonialdehyde (MDA) and PCR analyses (ICAM and Cox-1).
