RESUMO:

    Particulate systems chitosan (QUIT) and guar gum (GG) to transfer and colonic controlled release of triamcinolone acetonide (TRI) were prepared using simple coacervation technique with addition of sodium sulfate followed by drying by spray-drying method. With inlet temperature of 140° C and outlet 90° C, blower 6.5 and atomizing air 0.1 ppm. Different drug-polymer ratios (1:5, 1:10 and 1:20) and polymer-polymer (50:50, 25:75 and 75:25 w/w) were studied in terms of yield (%) encapsulation efficiency , EE (%) and loading (mg TRI/g SP), to optimize the preparation conditions of particulate systems obtained. Particulate systems with higher ratio of chitosan with regard to gum (75:25) showed better results in yield (%), however, as the EE (%) and loading showed no important differences among the other reasons (25:75) and (75:25) evaluated. The infrared spectrophotometry indicated the chemical and physical interactions between the polymers and promoted drug in different ratios and in the presence of triamcinolone particulate systems. The results obtained by the thermal analysis showed that a larger amount of chitosan improves the thermal stability of the systems. From the test in vivo chose to use the particulate systems in percentage ratio polymer:polymer 50:50 (%) for studies swelling and kinetics, as the relative amount of gum is important because of its swellable and mucoadhesive property for colonic release as well as chitosan, triamcinolone using the ratio w/w drug:polymer 1:10 as presented satisfactory results encapsulation (EE and loading).
