RESUMO:
Due to the growing interest and applications of biotechnological products, new biomolecule recovery and purification processes have been developed. In this scenario, ALE has attracted interest, since it presents advantages over traditional methods of separation because it is an integrative technique, combining the steps of clarification, concentration and purification in a single operation. Currently, the market provides chromatographic adsorbents for use with ALE, exploring different techniques, such as commercially known adsorbents like Streamline®.However, the search for new and better adsorbents for use in ALE is constant, mainly to minimize the effects of mass transfer. Thus, a judicious choice is required in the choice of resin as it represents a very important factor in the recovery and purification steps. Some parameters such as adsorbent density, size, type of binder and adsorption capacity are of utmost importance and should always be evaluated and improved when developing such adsorbents for use in ALE. Thus, the synthesis of suitable solid adsorbents has been highlighted in the studies developed in the last years since this element is essential in adsorption in expanded bed. The use of film adsorbents is presented as a suitable strategy to achieve high efficiency in ALE processes. Therefore, the present work aims to evaluate and optimize the conditions for the synthesis of a pellicular adsorbent from zirconia-yttria beads, coated with a layer of porous polymeric material (agarose), as well as to apply this resin to recover biomolecules in process of adsorption in expanded bed.
