SUMMARY:

The species Bryophyllum pinnatum (Lam) Oken has flavonoids, sugars and other bioactive compounds in its chemical constitution that are commonly used in cosmetic formulations with antioxidant, moisturizing and anti-aging properties. There is a global trend in the cosmetics sector in the search for products developed with raw materials of natural origin, in addition to being a very dynamic market, always looking for innovations. The objective of this work was to develop cosmetic formulations with moisturizing and antioxidant activities to prevent signs of skin aging, added with Bryophyllum pinnatum extract, and to evaluate the clinical moisturizing and anti-aging efficacy of the developed formulations. Therefore, the extract was chemically characterized in terms of sugar, phenolic compounds and total flavonoids. Safety was analyzed in vitro by cell viability tests and in vivo by skin compatibility studies in volunteers. The antioxidant action was evaluated in vitro using different methodologies and in vivo using Caenorhabditis elegans as a model organism. Antityrosinase activity was verified by enzymatic method. The formulations were developed based on the study of hydrophilic-lipophilic balance using the emulsion inversion point and ultrasonication methods, and subjected to preliminary and accelerated stability. A highperformance liquid chromatography method was developed and validated to quantify the marker named Bp1 in nanoemulsions. The extract featured flavonoids, phenolic compounds and sugars and is safe for topical use based on in vitro tests indicating non-cytotoxic nature and in vivo skin compatibility tests, in addition to supporting the “dermatologically tested” claim. It also acted on different stages of the oxidative cascade, and in vivo at the concentrations evaluated. The extract has also been shown to have antityrosinase activity. The developed formulations were stable for 30 days under different conditions, and the dosage of the Bp1 marker demonstrated chemical stability in the nanoemulsions after 10 and 30 days at temperatures of 4°C, 25 and

45°C. Thus, this work brings scientific contributions and technological innovation to the cosmetic sector, associated with the benefit of adding value to the plant's production chain, aiming to enhance social aspects, boost the bioeconomy and Brazilian biodiversity.
