SUMMARY:
Plants with therapeutic potential such as Bixa Orellana L. have been widely used in
research aimed at developing low-cost herbal medicines, however, before they can be
used, prior tests must be carried out. Toxicity tests guarantee the safety of future patients
in addition to being the first test to be carried out when therapeutic potential is found in a
species. This study aims to prepare isolated Nanostructured Lipid Carriers (CLN) with
nanoencapsulated annatto oil, characterize the formulations obtained and evaluate acute
oral toxicity. The formulations were prepared using the already standardized melting
emulsification method and then characterization was carried out to determine the success
of the formulations and their stability. Stability was established through mean diameter
(Z-average), polydispersity index (PdI) and zeta potential. Average diameter values of
less than 200nm determine that the particle sizes obtained are within the gauge scale. The
PdI values were below 0.2, which gives the formulations little polydisperse systems,
therefore being homogeneous formulations. The zeta potential demonstrated a value of -
17, this value may be related to the composition of the sample, as it has a stabilizer that
allows for the construction of a more electrostatically controlled system. The acute oral
toxicity test was carried out using a single dose of 2000 mg/Kg (mg.kg-1) of CLN, annatto
oil and CLN + nanoencapsulated annatto oil. Behavioral analyses, body weight, organ
weight, hematological and biochemical analyzes did not demonstrate toxicity. The results
obtained demonstrated that the formulations of CLN and CLN with nanoencapsulated
annatto oil remained stable during a period of 60 days, ensuring success in the
encapsulation process, and did not prove to be toxic.
