SUMMARY:
The present research aimed to develop an oral antiseptic lozenge containing
cetylpyridinium chloride (CPC), using direct compression technology. Therefore, the
analysis of pharmaceutical excipients and the evaluation of the characteristics of the
formulations were carried out. A preformulation study was carried out to assess the
compatibility of the active pharmaceutical ingredient (API) with the excipients using
thermoanalytical techniques: differential scanning calorimetry (DSC), thermogravimetry
(TG) and differential thermal analysis (DTA); and mid-infrared (FTIR) and near-infrared
(FT-NIR) spectroscopy. The pharmacotechnical characteristics of the mixture of the
components in powder form and the physicochemical and organoleptic characteristics of
the tablets obtained by direct compression were also evaluated. Preliminary and
accelerated stability studies were carried out under different conditions of storage and
packaging material. The DSC and TG results points to interactions between API and
menthol, with change in the melting point of API. The data obtained by FTIR and FT-NIR
demonstrated a physical interaction proven through statistical analysis of Pearson's
correlation. A product with adequate pharmacotechnical characteristics in terms of flow
and compressibility was obtained, considering the evaluations of characteristics of Carr
Index (CI), Hausner Ratio (RH), particle size distribution, flow velocity, angle of repose
and humidity. The lozenges obtained after direct compression met the characteristics,
considering weight, hardness, thickness, friability, disintegration, organoleptic
characteristics and pH value for evaluation. Stability studies showed that the product is
stable under the conditions studied when stored in glass or polyethylene terephthalate
(PET) bottles. Thus, it can be considered that CPC lozenges with adequate
pharmacotechnical characteristics and stability were obtained.
